Specificity and Use

SMC1A / SMC1 antibody was raised against phosphorylated synthetic peptide corresponding to a region within human SMC1 (locus link ID 5884) around serine 957. ( pSer957 ).
Recognizes human structural maintenance of chromosomes-1 (SMC1) which is phosphorylated at Ser957. SMC1 forms a heterodimer with SMC3, and is involved in sister chromatid cohesion and in DNA recombination and repair. SMC1 can be phosphorylated by ATM on serines 957 and 966 in vitro and in vivo in response to ionizing radiation. SMC1 also acts as a downstream effector in the ATM/NBS1 pathway of S-phase checkpoint.
Western Blot: LS-C43029 detects a band of approximately 150 kD in Hela cell lysates following treatment with 10 Gy ionizing radiation.

Proper cohesion of sister chromatids is a prerequisite for the correct segregation of chromosomes during cell division. The cohesin multiprotein complex is required for sister chromatid cohesion. This complex is composed partly of two structural maintenance of chromosomes (SMC) proteins, SMC3 and either SMC1B or the protein encoded by this gene. Most of the cohesin complexes dissociate from the chromosomes before mitosis, although those complexes at the kinetochore remain.

Lysate was subsequently treated with calf alkaline phosphatase (CIP) of left untreated as indicated by Western Blotting with Rabbit anti-phospho-SMC1 (pSER957) (LS-C43029).
