Immunofluorescent analysis of SNF2H staining in HeLa cells. Formalin-fixed cells were permeabilized with 0.1% Triton X-100 in TBS for 5-10 minutes and blocked with 3% BSA-PBS for 30 minutes at room temperature. Cells were probed with the primary antibody in 3% BSA-PBS and incubated overnight at 4 deg C in a humidified chamber. Cells were washed with PBST and incubated with a DyLight 594-conjugated secondary antibody (red) in PBS at room temperature in the dark.

Specificity and Use

SMARCA5 / SNF2H antibody was raised against kLH-conjugated, synthetic peptide corresponding to aa58-77 (MEEIFDDASPGKQKEIQEPD) of human hSNF2H (SWI/SNF-related matrix-associated actin-dependent chromatin a5). The immunizing sequence has 18/20 identical aa in rat and 17/20 identical aa in mouse. Percent identity by BLAST analysis: Human (100%); Gibbon, Monkey, Elephant, Dog, Bat, Horse, Pig (95%); Gorilla, Marmoset, Rat, Bovine, Rabbit, Opossum (90%); Mouse (85%); Chicken (80%).
Recognizes human SNF2H, Mr 120kD. A non-specific protein was also detected, Mr 90kD.
Suitable for use in Western Blot, Immunoprecipitation and Chromatin IP. Western Blot Analysis: A 1:1000-1:2000 dilution of this lot detected hSNF2H in nuclear extracts from HeLa cells. Immunoprecipitation: Has been reported by an independent laboratory to immunoprecipitate hSNF2H. Chromatin Immunoprecipitation: Has been reported by an independent laboratory to immunoprecipitate chromatin.

Helicase that possesses intrinsic ATP-dependent nucleosome-remodeling activity. Complexes containing SMARCA5 are capable of forming ordered nucleosome arrays on chromatin; this may require intact histone H4 tails. Also required for replication of pericentric heterochromatin in S-phase specifically in conjunction with BAZ1A. Probably plays a role in repression of polI dependent transcription of the rDNA locus, through the recruitment of the SIN3/HDAC1 corepressor complex to the rDNA promoter.
