Specificity and Use
SMAD1+5+9 antibody was raised against two synthetic peptides corresponding to amino acids 18-32 (LGWKQGDEEEKWAEK) and amino acids 315-330 (SNVNRNSTIENTRRHI) of human SMAD1;GenBank Accession No. AAP36050.1. They are 100% conserved or highly conserved between human and multiple species. Percent identity by BLAST analysis: Human, Gorilla, Orangutan, Gibbon, Marmoset, Mouse, Rat, Hamster, Elephant, Bat, Bovine, Rabbit, Pig, Turkey, Chicken, Lizard, Xenopus, Salmon, Stickleback, Pufferfish, Zebrafish, Drosophila (100%).
In A549, a 50 kD band is observed. The two synthetic peptide immunogens (LGWKQGDEEEKWAEK) and (SNVNRNSTIENTRRHI) used to make this are 100% conserved between SMAD1/5/8/9. Therefore, recognizes SMADs 1, 5, 8, and 9. may cross-react with SMAD2 or SMAD3. The actual SMADs detected with this may vary according to cell or tissue-type specific expression.
