Specificity and Use
SLC34A1/2 antibody was raised against human Type-IIa, NPT2/Napi-3 is ~640 aa (chromosome 8) and mouse and rat 637 aa proteins. A 20 AA synthetic peptide within the 2nd extracellular loop (between TM3-4) of Human NPT2 (206-225 aa) was synthesized, conjugated to KLH.
Recognizes human NaPi-2A. Species sequence homology: rat, mouse, sheep-100%, opossum-95% and chicken, zebra fish, frog, zebra fish-90%. The sequence is also highly conserved (~90%) in mouse, human and other species NPT2b/Type-Iib. Therefore, antibodies are expected to detect both Type2a and Type2b NPT2. No significant sequence homology is observed with other Pi-transporters (Type-I, III, or IV) transporters.
Suitable for use in ELISA and Western Blot. Western Blot: 1:1000-1:5000. Using chemiluminescence. Apparent molecular weight is 80-90kD and under reducing conditions 2 bands of 40-50kD are observed. May also form multimeric structures (170 and 200 kD). ELISA: 1:10000-1:50000. Control peptide can be used to coat ELISA plates at 1 ug/ml.
