SERPINA1 / Alpha 1 Antitrypsin Antibody - Immunofluorescent analysis of Alpha1-Antitrypsin was performed using 70% confluent log phase Hep G2 cells. The cells were fixed with 4% paraformaldehyde for 10 minutes, permeabilized with 0.1% Triton X-100 for 10 minutes, and blocked with 1% BSA for 1 hour at room temperature. The cells were labeled with Alpha1-Antitrypsin Mouse Monoclonal Antibody at 2 µg/mL in 0.1% BSA and incubated for 3 hours at room temperature and then labeled with Goat anti-Mouse IgG (H+L) Superclonal Secondary Antibody, Alexa Fluor® 488 conjugate a dilution of 1:2000 for 45 minutes at room temperature (Panel a: green). Nuclei (Panel b: blue) were stained with DAPI. F-actin (Panel c: red) was stained with Alexa Fluor® 555 Rhodamine Phalloidin. Panel d represents the merged image showing cytoplasmic localization. Panel e shows the no primary antibody control. The images were captured at 60X magnification.
SERPINA1 / Alpha 1 Antitrypsin Antibody - Western blot analysis was performed on membrane enriched extracts (30 ug) of (Fig 1) HT-29 (Lane 1) and Hep G2 (Lane 2). Likewise, Western blot analysis was performed on (Fig 2) condition media of Hep G2 (Lane 1).The blots were probed with Anti-Alpha1-Antitrypsin Mouse Monoclonal Antibody and detected by chemiluminescence using Goat anti-Mouse IgG (H+L) Superclonal Secondary Antibody, HRP conjugate.A~55 kDa band corresponding Alpha1-Antitrypsin was observed in HT-29, Hep G2 and in Hep G2 conditioned media. The observed shift in molecular weight from 50 kDa is due to glycosylation of the protein. Known quantity of protein samples were electrophoresed using4-12 % Bis-Tris gel. Resolved proteins were then transferred onto a nitrocellulose membrane. The membrane was probed with the relevant primary and secondary Antibody using iBind Flex Western Starter Kit.
