  • Work with LifeSpan to design a custom immunohistochemistry to address your specific biological question. Outsource the entire localization process without having to worry about finding and characterizing target specific antibodies, sourcing and validating difficult-to-find tissues, and having the ability to interpret the resulting immunostaining in relation to complex human pathologies.

  • TCR Screening Services

  • Test your therapeutic antibodies in immunohistochemistry against a broad panel of normal frozen human tissue types in order to determine potential unintended binding. Our non-GLP TCR services are designed on the FDA recommendation outlined in their "Points to Consider in the Manufacture and Testing of Monoclonal Antibody Products for Human Use".

Specificity and Use

RBL2 / RB2 p130 antibody was raised against this affinity purified antibody was prepared by repeated immunizations with a synthetic peptide corresponding to the Spa310 sequence of pRb2/p130 protein. A residue of cysteine was added to facilitate coupling.
This antiserum is directed against Spa310 and reacts with the Spa3120 domain of pRb2/p130 from human tissues. Based on the sequence we expect this antibody to react as well Spa310 from chimpanzee and orangutan, and with lesser affinity, Spa310 from horse, dog, bovine, rat and opossum.
This affinity purified antibody is suitable for ELISA and western blotting. Specific conditions for reactivity should be optimized by the end user. Expect a band approximately 130 kD in size corresponding to pRb2/p130, and a band approximately 4.2kD in size corresponding to Spa310 peptide (latter not shown), by western blotting in the appropriate cell lysate or extract.

0.02 M potassium phosphate, 0.15 M sodium chloride, pH 7.2, 0.1% sodium azide, sterile filtered
Short term 4°C, long term aliquot and store at -20°C, avoid freeze thaw cycles.

About RBL2 / RB2 p130

Q08999 NM_005611 NP_005602.3

RBL2 Antibody, p130 Antibody, Retinoblastoma-like 2 (p130) Antibody, RBR-2 Antibody, Rb2 Antibody, Retinoblastoma-like protein 2 Antibody

Key regulator of entry into cell division. Directly involved in heterochromatin formation by maintaining overall chromatin structure and, in particular, that of constitutive heterochromatin by stabilizing histone methylation. Recruits and targets histone methyltransferases SUV420H1 and SUV420H2, leading to epigenetic transcriptional repression. Controls histone H4 'Lys-20' trimethylation. Probably acts as a transcription repressor by recruiting chromatin-modifying enzymes to promoters.

Western blot

Western blot
Anti-Spa310 Antibody - Western Blot. Western blot of affinity purified anti-Spa310 antibody shows detection of endogenous pRb2/p130 protein in whole LNCaP cell extracts. The band at ~130 kD, indicated by the arrowhead, corresponds to the expected molecular weight of pRb2/p130. The membrane was blocked overnight with a milk buffer at 4C. Primary antibody was diluted 1:500 and reacted with the membrane overnight at 4C. ECL was used for detection.
