Transcription factor implicated in the cell fate determination in various organs. Binds to the E-box consensus sequence 5'-CANNTG-3'. Plays a role in early and late pancreas development and differentiation. Important for determining whether cells allocated to the pancreatic buds continue towards pancreatic organogenesis or revert back to duodenal fates. May be involved in the maintenance of exocrine pancreas-specific gene expression including ELA1 and amylase.
PTF1A in Human Pancreas. PTF1A was detected in immersion fixed paraffin-embedded sections of human pancreas using Goat Anti-Human PTF1A Antigen Affinity-purified Polyclonal Antibody at 15 ug/ml overnight at 4°C. Tissue was stained using Anti-Goat HRP-DAB (brown) and counterstained with hematoxylin (blue). Specific staining was localized to nuclei.
PTF1A in Human Pancreatic Cancer Cells. PTF1A was detected in immersion fixed human pancreatic cancer cells using Goat Anti-Human PTF1A Antigen Affinity-purified Polyclonal Antibody at 10 ug/ml for 3 hours at room temperature. Cells were stained using NorthernLights 557-conjugated Anti-Goat IgG Secondary Antibody (red, upper panel) and counterstained with DAPI (blue, lower panel). Specific staining was localized to nuclei.
Detection of Human PTF1A by Western Blot. Western blot shows lysates of Jurkat human acute T cell leukemia cell line. PVDF Membrane was probed with 1 ug/ml of Goat Anti-HumanPTF1A Antigen Affinity-purified Polyclonal Antibody followed by HRP-conjugated Anti Goat IgG Secondary Antibody. A specific band was detected for PTF1A at approximately 45 kD (as indicated). This experiment was conducted under reducing conditions.
