Services
  • Immunohistochemistry Services

  • Work with LifeSpan to design a custom immunohistochemistry to address your specific biological question. Outsource the entire localization process without having to worry about finding and characterizing target specific antibodies, sourcing and validating difficult-to-find tissues, and having the ability to interpret the resulting immunostaining in relation to complex human pathologies.

  • TCR Screening Services

  • Test your therapeutic antibodies in immunohistochemistry against a broad panel of normal frozen human tissue types in order to determine potential unintended binding. Our non-GLP TCR services are designed on the FDA recommendation outlined in their "Points to Consider in the Manufacture and Testing of Monoclonal Antibody Products for Human Use".

Immunohistochemical analysis of PSMB8 staining in rat heart formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with hematoxylin and mounted with DPX.

Specificity and Use

PSMB8 / LMP7 antibody was raised against synthetic peptide C-DVSDLLHQYREANQ from the C-terminus of human PSMB8 (NP_004150.1; NP_683720.2). Percent identity by BLAST analysis: Human, Chimpanzee, Gorilla, Monkey (100%); Gibbon, Marmoset, Elephant (93%); Goat, Panda, Bat, Bovine, Horse, Pig (86%).
LS-E27091 - Lyophilized - 100 µg - $145.00
Immunizing peptide used to generate LS-B9206. Useful for pre-absorption and neutralization of the antibody's antigen binding site.
aa263-276
Human PSMB8. This antibody is expected to recognize both reported isoforms (NP_004150.1; NP_683720.2).
Peptide ELISA: antibody detection limit dilution 1:8000. Western blot: Approx 22kDa band observed in Human Liver, Lung and Lymph Node lysates (calculated MW of 29.8kDa according to precursor NP_004150.1). The observed molecular weight corresponds to earlier findings in literature with different antibodies (Schwarz et al, J Immunol. 2000 Jul 15;165(2):768-78. PMID: 10878350. Recommended concentration: 0.01-0.03 ug/ml.

The proteasome is a multicatalytic proteinase complex with a highly ordered ring-shaped 20S core structure. The core structure is composed of 4 rings of 28 non-identical subunits; 2 rings are composed of 7 alpha subunits and 2 rings are composed of 7 beta subunits. Proteasomes are distributed throughout eukaryotic cells at a high concentration and cleave peptides in an ATP/ubiquitin-dependent process in a non-lysosomal pathway.
