Specificity and Use

PSMA1 antibody was raised against synthetic Peptide: CP(249)ADEPAEKADEPMEH(263). Immunizing peptide corresponds to amino acid residues 249-263 from the C-terminus of human proteasome 20S C2 subunit. This sequence is 93% conserved in the rat and chicken protein and 87% conserved in the mouse protein. Percent identity by BLAST analysis: Human, Gorilla, Orangutan, Gibbon, Monkey, Marmoset, Rat, Hamster, Porcine, Dog, Bat, Bovine, Horse, Rabbit, Opossum (100%); Mouse, Elephant, Turkey, Chicken (93%); Platypus (86%).

Detects proteasome 20S C2 subunit. Species cross-reactivity: Human, Mouse, Rat, canine and Hamster. Expected to cross-react with Chicken due to sequence homology.
Suitable for use in ELISA and Western Blot. Western Blot: 1 ug/ml detects a 29kD protein representing Proteasome 20S C2 subunit from rat brain extract.

The proteasome is a multicatalytic proteinase complex with a highly ordered ring-shaped 20S core structure. The core structure is composed of 4 rings of 28 non-identical subunits; 2 rings are composed of 7 alpha subunits and 2 rings are composed of 7 beta subunits. Proteasomes are distributed throughout eukaryotic cells at a high concentration and cleave peptides in an ATP/ubiquitin-dependent process in a non-lysosomal pathway.
