  • Work with LifeSpan to design a custom immunohistochemistry to address your specific biological question. Outsource the entire localization process without having to worry about finding and characterizing target specific antibodies, sourcing and validating difficult-to-find tissues, and having the ability to interpret the resulting immunostaining in relation to complex human pathologies.

  • TCR Screening Services

  • Test your therapeutic antibodies in immunohistochemistry against a broad panel of normal frozen human tissue types in order to determine potential unintended binding. Our non-GLP TCR services are designed on the FDA recommendation outlined in their "Points to Consider in the Manufacture and Testing of Monoclonal Antibody Products for Human Use".

Specificity and Use

PRSS3 / Trypsin 3 antibody was raised against nS0-derived recombinant human rhTrypsin-3.
Recognizes both pro and mature forms of human trypsin 3. Species sequence homology: chimpanzee: 96%, rat: 76%, mouse: 74%, dog: 72%. and chicken: 67%. Does not cross-react with rhTrypsin-1 or rhTrypsin-2.
Suitable for use in Neutralization and Western Blot. Neutralization: The antibody was preincubated with the enzyme at different molar ratios at RT for 30 minutes. The enzyme was then assayed with a fluorogenic peptide substrate. 50% of the proteolytic activity was inhibited by the antibody at approximately 0.15 ug/ml (IC 50) under conditions in which the enzyme was present at 0.03 ug/ml and the substrate concentration was 10uM. Considering the molecular masses of the enzyme (26kD) and the antibody (150kD), IC 50 was achieved at approximately 1:1 molar ratio of the antibody to the enzyme. Western blot:1-2 ug/ml. Using a colorimetric detection system, the detection limit for rhTrypsin-3 is ~25 ng/lane under non-reducing conditions. Chemiluminescent detection will increase sensitivity by 5 to 50 fold. Reducing conditions are not recommended.

PRSS3 / Trypsin 3 is a trypsinogen, which is a member of the trypsin family of serine proteases. This enzyme is expressed in the brain and pancreas and is resistant to common trypsin inhibitors. It is active on peptide linkages involving the carboxyl group of lysine or arginine. This gene is localized to the locus of T cell receptor beta variable orphans on chromosome 9. Four transcript variants encoding different isoforms have been described for this gene.
