Specificity and Use

PRKDC / DNA-PKcs antibody was raised against human DNA-dependent protein kinase purified from HeLa cells. Cellular Localization: Nuclear.
Recognizes the catalytic subunit of DNA-dependent protein kinase (DNA-PKcs) at 460kD. Species cross-reactivity: human, mouse, hamster, Xenopus and chicken.
Suitable for use in Gel Supershift, Immunohistochemistry, Immunofluorescence, Immunoprecipitation and Western Blot. Immunoprecipitation (Native and denatured): 2 ug/mg protein lysate (Use Protein G). Western Blot: 1-2 ug/ml. Immunohistochemistry (Formalin/paraffin): 2-4 ug/ml for 30 min at RT. Note: Staining of formalin-fixed tissues requires boiling tissue sections in 10 mM citrate buffer, pH 6 for 10-20minutes followed by cooling at RT for 20 minutes. Gel Supershift: Use antibody at 1 mg/ml. Positive control: LS174T cells. Tonsil or testis.

Serine/threonine-protein kinase that acts as a molecular sensor for DNA damage. Involved in DNA non-homologous end joining (NHEJ) required for double-strand break (DSB) repair and V(D)J recombination. Must be bound to DNA to express its catalytic properties. Promotes processing of hairpin DNA structures in V(D)J recombination by activation of the hairpin endonuclease artemis (DCLRE1C). The assembly of the DNA-PK complex at DNA ends is also required for the NHEJ ligation step.
