Specificity and Use

PR / Progesterone Receptor antibody was raised against pgR from a human endometrial carcinoma (EnCa 101) grown in athymic mice. Cellular Localization: Nuclear.
Equally recognizes two proteins: 116kD (triplet) and 81kD (singlet), identified as the hormone-binding high (B) and low (A) MW forms of human PgR. Species cross-reactivity: Human, equine, sheep and porcine.
Suitable for use in Immunofluorescence, Immunoprecipitation, Western Blot, Immunohistochemistry and Sedimentation in Sucrose Gradient. Immunoprecipitation (Use Protein G): 2 ug/mg protein lysate. Western Blot: 1-2 ug/ml for 2hrs at RT. Immunohistochemistry (Formalin/paraffin): 1:50-1:100 for 60 min at RT. Staining of formalin/paraffin tissues requires boiling tissue sections in 10 mM citrate buffer, pH 6.0 for 10-20 min followed by cooling at RT for 20 min. Positive control: T47D or breast Carcinoma.

The progesterone receptor, a NR3 Steroid Receptor, is induced in granulosa cells of preovulatory follicles in response to surges in luteinizing hormone. It is essential for ovulation and egg implantation. A decrease in expression of endometrial progesterone receptor results in infertility in women. This receptor mediates the progesterone-induced transcription of proteases including ADAMTS1 and cathepsin L.
