Specificity and Use

E. coli-derived rhPlexin C1 (aa49-161).

Selected for its ability to recognize human Plexin C1 in the applications listed below. In direct ELISAs and Western Blots, this antibody shows ~10% cross-reactivity with rmPlexin A1, rmPlexin A2, rmPlexin A3, rhPlexin B1, and less than 1% cross-reactivity with rhPlexin D1.
Western Blot: This antibody can be used at 0.1-0.2 ug/ml with the appropriate secondary reagents to detect human Plexin C1. The detection limit for rhPlexin C1 is approximately 1 ng/lane under non-reducing and reducing conditions. Flow Cytometry: This antibody has been tested on white blood monocytes for use in flow cytometry. Dilute this antibody to 50 ug/ml and add 10 ul of the diluted solution to 1-2.5 x 10^5 cells in a total reaction volume not exceeding 200 L. The binding of unlabeled antibodies may be visualized by adding 10 ul of a 25 ug/ml stock solution of a secondary developing reagent such as anti-goat lgG conjugated to a fluorochrome. Direct ELISA: This antibody can be used at 0.5-1.0 ug/ml with the appropriate secondary reagents to detect human Plexin C1. The detection limit for rhPlexin C1 is approximately 0.5 ng/well.

PLXNC1 / Plexin C1 is a member of the plexin family. Plexins are transmembrane receptors for semaphorins, a large family of proteins that regulate axon guidance, cell motility and migration, and the immune response. The encoded protein and its ligand regulate melanocyte adhesion, and viral semaphorins may modulate the immune response by binding to this receptor. The encoded protein may be a tumor suppressor protein for melanoma.
