PECAM-1 / CD31 Antibody - Western blot analysis of CD31 using anti-CD31 antibody. Electrophoresis was performed on a 5-20% SDS-PAGE gel at 70V (Stacking gel) / 90V (Resolving gel) for 2-3 hours. The sample well of each lane was loaded with 50ug of sample under reducing conditions. lane 1: HELA whole cell lysate, lane 2: U937 whole cell lysate, lane 3: MM231 whole cell lysate, lane 4: JURKAT whole cell lysate, lane 5: RAJI whole cell lysate. After Electrophoresis, proteins were transferred to a Nitrocellulose membrane at 150mA for 50-90 minutes. Blocked the membrane with 5% Non-fat Milk/ TBS for 1.5 hour at RT. The membrane was incubated with rabbit anti-CD31 antigen affinity purified polyclonal antibody at 0.5 µg/mL overnight at 4°C, then washed with TBS-0.1% Tween 3 times with 5 minutes each and probed with a goat anti-rabbit IgG-HRP secondary antibody at a dilution of 1:10000 for 1.5 hour at RT. The signal is developed using an Enhanced Chemiluminescent detection (ECL) kit with Tanon 5200 system. A specific band was detected for CD31 at approximately 82KD. The expected band size for CD31 is at 82KD.

PECAM-1 / CD31 Antibody - Flow Cytometry analysis of U937 cells using anti-CD31 antibody. Overlay histogram showing U937 cells stained with anti-CD31 antibody (Blue line). The cells were blocked with 10% normal goat serum. And then incubated with rabbit anti-CD31 Antibody (1µg/1x106 cells) for 30 min at 20°C. DyLight®488 conjugated goat anti-rabbit IgG (5-10µg/1x106 cells) was used as secondary antibody for 30 minutes at 20°C. Isotype control antibody (Green line) was rabbit IgG (1µg/1x106) used under the same conditions. Unlabelled sample (Red line) was also used as a control.

E.coli-derived human CD31 recombinant protein (Position: Q28-G382). Human CD31 shares 65% and 68% amino acid (aa) sequences identity with mouse and rat CD31, respectively.

Specificity

Expressed on platelets and leukocytes and is primarily concentrated at the borders between endothelial cells. Isoform Long predominates in all tissues examined. Isoform Delta12 is detected only in trachea. Isoform Delta14-15 is only detected in lung. Isoform Delta14 is detected in all tissues examined with the strongest expression in heart. Isoform Delta15 is expressed in brain, testis, ovary, cell surface of platelets, human umbilical vein endothelial cells (HUVECs), Jurkat T-cell leukemia, human erythroleukemia (HEL) and U-937 histiocytic lymphoma cell lines (at protein level). .
