Specificity and Use
PARP4 / VPARP antibody was raised against minor vault p193 protein (VPARP; PARP-4).
Recognizes epitope aa 506-510 (VALGK) of human minor vault p193 protein.
Immunohistochemistry: Use 1:10-1:20 dilution for staining of paraformaldehyde (4%) fixed cytospin preparations and frozen tissue sections. Apply the following pretreatment: 10 min in 20 mM glycine in PBS (pH 7.5) and 10 min in 6N guanidine hydrochloride in 50 mM TRIS-HCl (pH 7.5); Schroeijers et al., 2000. Western Blot: Use 1:10-1:20 dilution (ECL).
PARP4 / VPARP encodes poly(ADP-ribosyl)transferase-like 1 protein, which is capable of catalyzing a poly(ADP-ribosyl)ation reaction. This protein has a catalytic domain which is homologous to that of poly (ADP-ribosyl) transferase, but lacks an N-terminal DNA binding domain which activates the C-terminal catalytic domain of poly (ADP-ribosyl) transferase.
