Specificity and Use

PARP1 antibody was raised against purified bovine PARP-1 (poly(ADP-ribose) polymerase-1).
Recognizes human and bovine PARP-1. The epitope is in the zinc finger II DNA binding domain of PARP-1. Detects bands of ~116kD (intact PARP-1) and ~24kD (apoptosis-induced cleavage fragment) by Western blot. Does not cross-react with mouse or rat PARP-1.
ELISA: >1:200000. Dilution buffer: PBS, pH 7.4, containing 1% non fat dried milk and 0.05% Tween 20. Detectable antigen range: 50 ng of PARP-1. Immunocytochemistry: 1:500. Dilution buffer: 50 mM TRIS, pH 7.5, containing 150 mM NaCl and 5% milk. Immunoprecipitation: 1:50. Dilution buffer: RiPA buffer Protein G-Sepharose. Western Blot: 1:5000. See also separate Western blot protocol. Functional Application: Neutralizes the biological activity of PARP (~80%).

Mediates the poly(ADP-ribosyl)ation of APLF and CHFR.
