Specificity and Use

Osteonectin / SPARC antibody was raised against mouse myeloma cell line NS0-derived recombinant human SPARC/Osteonectin Ala18-Ile303 Accession # P09486.

Detects human SPARC/Osteonectin in direct ELISAs and Western blots. In direct ELISAs, approximately 25% cross-reactivity with.
Western Blot, 1 µg/ml. Immunohistochemistry, 5-15 µg/ml, Intracellular Staining by Flow Cytometry, 2.5 µg/106 cells, MG-63 human osteosarcoma cell line fixed with paraformaldehyde and permeabilized with. saponin.

SPARC (secreted protein acidic and rich in cysteine)/Osteonectin is a matricellular glycoprotein that modulates cellular interactions with the ECM and is expressed in tissues undergoing remodeling. It functions as a de-adhesive protein, as a modulator of growth factor activity, and as a cell-cycle inhibitor. It induces changes in endothelial cell shape via F-actin, coincident with the appearance of intercellular gaps, that provide a paracellular pathway for extravasation of macromolecules.

SPARC/Osteonectin in Human Ovary. SPARC/Osteonectin was detected in immersion fixed paraffin-embedded sections of human ovary using Goat Anti-Human SPARC/Osteonectin Antigen Affinity-purified Polyclonal Antibody at 3 ug/ml overnight at 4°C. Tissue was stained using Anti-Goat HRP-DAB (brown) and counterstained with hematoxylin (blue).

Detection of Human SPARC by Western Blot. Western blot shows lysates of human kidney (medulla) tissue. PVDF membrane was probed with 1 ug/ml of Goat Anti-Human SPARC/Osteonectin Antigen Affinity purified Polyclonal Antibody followed by HRP-conjugated Anti-Goat IgG Secondary Antibody. A specific band was detected for SPARC at approximately 43 kD (as indicated). This experiment was conducted under reducing conditions.
