Specificity and Use

OGG1 antibody was raised against synthetic peptide C-HPKTSQAKGPSPLANK from an internal region of mouse OGG1 (NP_035087.2). Percent identity with other species by BLAST analysis: Mouse (100%), Rat (88%), Hamster (88%), Elephant (88%), Panda (88%), Bovine (88%), Human (81%), Gorilla (81%), Monkey (81%), Dog (81%), Horse (81%).

Immunohistochemistry: LS-B2399 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B2399 was determined to be 2.5 ug/ml.

OGG1 is the enzyme responsible for the excision of 8-oxoguanine, a mutagenic base byproduct which occurs as a result of exposure to reactive oxygen. The action of this enzyme includes lyase activity for chain cleavage. Alternative splicing of the C-terminal region of this gene classifies splice variants into two major groups, type 1 and type 2, depending on the last exon of the sequence. Type 1 alternative splice variants end with exon 7 and type 2 end with exon 8.
