Specificity and Use

NTRK1 / TrkA antibody was raised against nS0-derived rhTrkA extracellular domain.
Human TrkA.
Neutralization of Human TrkA bioactivity. The exact concentration of antibody required to neutralize cell surface TrkA mediated activity is dependent on cell type, growth conditions and the type of activity studied. The Neutralization Dose50 (ND50) for this antibody is defined as that concentration of antibody required to yield one-half maximal inhibition of the ligand-induced activity on a responsive cell line. The ND50 for this lot of anti-human TrkA antibody was determined to be approximately. 3-12 µg/ml. The P-NGF-dependent 3H-thymidine incorporation by TF-1 cells was used as an assay. Western blot-This antibody can be used at 0.1-0.2 µg/ml with the appropriate secondary reagents to detect human TrkA. The detection limit for rhTrkA is approximately 5 ng/lane under non-reducing and reducing conditions. Immunohistochemistry-This antibody will detect TrkA in cells and tissues. The working dilution is 15 µg/ml. Flow cytometry-This antibody can be used at 3-10 µg/ml/106 cells with an appropriate secondary antibody for indirect immunofluorescence staining of cells by flow cytometry. Direct ELISA-This antibody can be used at 0.5-1.0 µg/ml with the appropriate secondary reagents to detect human TrkA. The detection limit for rhTrkA is approximately 0.3 ng/well. In this format, this antibody shows approximately. 50% cross-reactivity with rrTrkA and 10% cross-reactivity with rhTrkB and rhTrKC.

Receptor tyrosine kinase involved in the development and the maturation of the central and peripheral nervous systems through regulation of proliferation, differentiation and survival of sympathetic and nervous neurons.
