Specificity and Use

NR5A2 / LRH-1 antibody was raised against synthetic 16 amino acid peptide from internal region of human NR5A2. Percent identity with other species by BLAST analysis: Human, Gorilla, Gibbon, Monkey, Marmoset, Mouse, Rat, Dog, Hamster, Elephant, Panda, Horse, Opossum, Turkey, Chicken, Platypus, Xenopus, Trout, Zebrafish, Stickleback (100%); Sheep, Bovine, Bat, Pufferfish (94%); Chimpanzee (88%); Catfish (81%).
Human NR5A2. BLAST analysis of the peptide immunogen showed no homology with other human proteins.
Immunohistochemistry: LS-A2453 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-A2453 was determined to be 2.5 - 5 ug/ml. Antibody does not work with tissues fixed in 4% PFA.

Alpha1-fetoprotein transcription factor (Ftf), a NR5 fushi tarazu-like receptor, also is known as 'human B1-binding factor' (HB1F), 'CYP7A promoter-binding factor' (CPF), and 'liver receptor homolog 1' (LRH1). Ftf has been shown to affect the transcription of hepatic genes including alpha fetoprotein encoding gene (AFP), viral hepatitis B enhancer II, and 7-alpha-hydroxylase encoding gene (CYP7A1). Ftf has also been shown to affect hepatic cholesterol metabolism and bile acid synthesis.
