Specificity and Use

NEFH / NF-H antibody was raised against native NF-H purified from pig spinal cord.
Clone NAP4 irecognizes phosphorylated NF-H KSP (lysine-serine-proline) type sequences. In some species there is some cross-reactivity with the related phosphorylated KSP sequences found in the related neurofilament subunit NF-M. The antibody recognizes NF-H strongly in all mammals tested to date and also in chicken. It recognizes neurofilaments in frozen sections in tissue culture and in formalin fixed sections.
The ascites solution has a high titre and can be used at dilutions of at least 1:1000 in immunofluorescence experiments. In western blotting using chemiluminescence it can be used at dilutions of 1:10000 or lower.

Neurofilaments are type IV intermediate filament heteropolymers composed of light, medium, and heavy chains. Neurofilaments comprise the axoskeleton and functionally maintain neuronal caliber. They may also play a role in intracellular transport to axons and dendrites. This gene encodes the heavy neurofilament protein. This protein is commonly used as a biomarker of neuronal damage and susceptibility to amyotrophic lateral sclerosis (ALS) has been associated with mutations in this gene.


Human cerebellar cortex fixed in formalin, embedded in paraffin and stained with NEFH / NF-H antibody using the ABC (avidin biotin conjugate) method. The section was counterstained with hematoxylin-Eosin (blue). NEFH / NF-H antibody stains prominent basket cell axons surrounding the large Purkinje neurons. Granule cell layer at bottom of image, molecular layer at top.
