NALP3 / NLRP3 is a pyrin-like protein containing a pyrin domain, a nucleotide-binding site (NBS) domain, and a leucine-rich repeat (LRR) motif. This protein interacts with the apoptosis-associated speck-like protein PYCARD/ASC, which contains a caspase recruitment domain, and is a member of the NALP3 inflammasome complex. This complex functions as an upstream activator of NF-kappaB signaling, and it plays a role in the regulation of inflammation, the immune response, and apoptosis.
NALP3 in THP-1 Human Cell Line. NALP3 was detected in immersion fixed THP-1 human acute monocytic leukemia cells, unstimulated (lower panel) and stimulated (upper panel) with PMA and LPS, using Sheep Anti-Human NALP3 Antigen Affinity-purified Polyclonal Antibody at 10 ug/ml for 3 hours at room temperature. Cells were stained using NorthernLights 557-conjugated Anti-Sheep IgG Secondary Antibody (red) and counterstained with DAPI (blue). Specific staining was localized to cytoplasm.
Detection of Human NALP3 by Western Blot. Western blot shows lysates of HEK293 human embryonic kidney cell line either mock transfected, transfected with human NALP1 or transfected with NALP3 and THP-1 human acute monocytic leukemia cell line untreated (-) or treated (+) with 20 ng/ml PMA for 72 hours and 1 ug/ml LPS for 24 hours. PVDF Membrane was probed with 0.5 ug/ml of Sheep Anti-Human NALP3 Antigen Affinity-purified Polyclonal Antibody followed by HRP-conjugated Anti-Sheep IgG Secondary Antibody. A specific band was detected for NALP3 at approximately 115 kD (as indicated). This experiment was conducted under reducing conditions.
