Specificity and Use

MPS1 / TTK antibody was raised against his-tagged fusion protein corresponding to aa3-856 of human Mps1, previously known at TTK/PYK.
C-Terminus
Recognizes human Mps1, Mr ~97kD. This antibody is known to inhibit the autophosphorylation/ kinase activity of Mps1. Does not react with mouse.
Suitable for use in Western Blot, Immunoprecipitation and Immunocytochemistry. Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system. Immunoprecipitation: Has been reported by an independent laboratory to immunoprecipitate Mps1 from HeLa total cell extract. Immunocytochemistry: Has been reported by an independent laboratory to show positive immunostaining for Mps1 in U2OS or HeLa cells permeabilized with 0.5% Triton-X-100 and fixed with methanol or paraformaldehyde.

0.1 M Tris-glycine, pH 7.4, 0.15 M sodium chloride, 0.05% sodium azide, 30% glycerol.

MPS1 / TTK is a dual specificity protein kinase with the ability to phosphorylate tyrosine, serine and threonine. Associated with cell proliferation, this protein is essential for chromosome alignment at the centromere during mitosis and is required for centrosome duplication. It has been found to be a critical mitotic checkpoint protein for accurate segregation of chromosomes during mitosis.
