Specificity and Use

MCL1 / MCL 1 antibody was raised against synthetic peptide corresponding to amino acids 121-139 of the Mcl-1 protein.
aa121-139
Recognizes the human Mcl-1 apoptosis related protein, a member of the Bcl-2 family. Western Blotting experiments show a band of approx 38kD corresponding to intact Mcl-1.
Suitable for use in Immunohistochemistry and Western Blot. Immunohistochemistry (paraffin): 1:200-1:1000. Note: Requires antigen retrieval using heat treatment prior to staining of paraffin sections. Optimal results may be obtained using Bouin's fixative and heat mediated antigen retrieval although neutral buffered formalin may also be used. Western Blot: 1:500-1:1000. Positive control: Human Tonsil.

Myeloid cell leukimia-1 (Mcl-1) is a member of the Bcl-2 family of proteins that can act to promote cell survival. While the mechanism by which Mcl-1 inhibits apoptosis is not known, it is thought that it may heterodimerize and neutralize pro-apoptotic members of the Bcl-2 family such as Bim or Bak. Mcl-1 was originally identified in differentiating myeloid cells, but has since been shown to be expressed in multiple cell types.
