MAP4K4 is a MSN protein kinase. The protein has a structure similar to other GCK (Group I) kinases and contains two consensus SH3 binding motifs, which have been shown to interact with the SH3 domains of the SH2/SH3 adapter protein Nck. NIK can be activated by tumor necrosis factor (TNF)-alpha. Activated NIK has been shown to activate c-Jun N-terminal kinase (JNK) and to bind and activate the plasma membrane Na-H exchanger NHE1.
