MAP3K14
mitogen-activated protein kinase kinase kinase 14
MAP3K14, also known as NIK for NF-kappaB-inducing kinase, is a MAP3K type protein kinase. NIK/MAP3K14 is a member of a NF-kappaB-inducing signalling cascade common to receptors of the tumor-necrosis/nerve-growth factor (TNF/NGF) family and to the interleukin-1 type-I receptor. The protein has been shown to mediate TNF activation of NF-kappaB by associating with the adapter protein TRAF2. NIK/MAP3K14 (-/-) mice lack lymph nodes and have defective development of Peyer patches. A mutation in the C-terminus of the protein has been shown to cause alymphoplasia, a disease characterized in part by the systemic absence of lymph nodes and Peyer patches, in aly/aly mice.
