  • Work with LifeSpan to design a custom immunohistochemistry to address your specific biological question. Outsource the entire localization process without having to worry about finding and characterizing target specific antibodies, sourcing and validating difficult-to-find tissues, and having the ability to interpret the resulting immunostaining in relation to complex human pathologies.

  • TCR Screening Services

  • Test your therapeutic antibodies in immunohistochemistry against a broad panel of normal frozen human tissue types in order to determine potential unintended binding. Our non-GLP TCR services are designed on the FDA recommendation outlined in their "Points to Consider in the Manufacture and Testing of Monoclonal Antibody Products for Human Use".

Specificity and Use

MAD2B / REV7 antibody was raised against synthetic peptide from human MAD2L2.
Aa 3-14 of Human MAD2L2. MAD2L2 is also known as MAD2B, REV7 and mitotic arrest deficient-like 2 protein.
Immunohistochemistry: LS-B390 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B390 was determined to be 10 ug/ml.

0.02 M potassium phosphate, 0.15 M sodium chloride, pH 7.2, 0.01% sodium azide.
Long term: -20°C; Short term: +4°C. Avoid repeat freeze-thaw cycles.

About MAD2B / REV7

Q9UI95 NM_006341 NP_006332.3

MAD2L2 Antibody, POLZ2 Antibody, REV7 homolog Antibody, REV7 Antibody, MAD2-like protein 2 Antibody, MAD2B Antibody, HREV7 Antibody

Adapter protein able to interact with different proteins and involved in different biological processes. Mediates the interaction between the error-prone DNA polymerase zeta catalytic subunit REV3L and the inserter polymerase REV1, thereby mediating the second polymerase switching in translesion DNA synthesis. Translesion DNA synthesis releases the replication blockade of replicative polymerases, stalled in presence of DNA lesions.

Anti-MAD2L2 antibody IHC of human brain, cerebellum. Immunohistochemistry of formalin-fixed, paraffin-embedded tissue after heat-induced antigen retrieval. Antibody LS-B390 concentration 10 ug/ml.

Western blot

Western blot
Anti-MAD2L2 Antibody - Western Blot. Affinity Purified Rabbit anti-MAD2L2 was used at a 1:500 dilution to detect human MAD2L2 by western blot. Both HeLa whole cell lysate (lane 1) and nuclear lysate (lane 2) were probed using this antibody. This antibody clearly detects a ~20 kD band corresponding to human MAD2L2 (predicted molecular weight is 24 kD). Approximately 20 ug of each lysate was loaded on a 10% SDS-PAGE. Primary antibody was reacted with the membrane at room temperature for 1 h. After subsequent washing, a 1:2000 dilution of HRP conjugated Gt-a-Rabbit IgG was used for visualization. Exposure time was 30 sec.
