Specificity and Use

Full length lactate dehydrogenase protein isolated from rabbit muscle.
Lactate Dehydrogenase [Rabbit Muscle]. BLAST analysis was used to determine that cross reactivity is suggested for both muscle and heart isoforms (LDH-A and LDH-B) from most mammalian species
This antibody has been tested for use in ELISA and by western blot. Specific conditions for reactivity should be optimized by the end user. Expect a band approximately 36 kD in size corresponding to LDH western blotting in the appropriate cell lysate or extract.

About LDH / Lactate Dehydrogenase

Lactate dehydrogenase is composed of four subunits (tetramer). The two most common subunits are the LDH-M and LDH-H protein, encoded by the LDHA and LDHB genes, respectively. These two subunits can form five possible tetramers (isoenzymes): 4H, 4M, and the three mixed tetramers (3H1M, 2H2M, 1H3M).


Lactate Dehydrogenase Antibody-Immunofluorescence. Biotin conjugated anti-lactate dehydrogenase antibody was used to stain HeLa cells by immunofluorescence. HeLa cells were plated in 12 well plates, fixed for 5 min in 1:1 Methanol: Acetone, blocked with MB-071 (preservative free) for 15 min and stained 1 hr with antibody 1:200 in blocking buffer. Plate was washed 3X in PBS, and primary biotin conjugate was detected by DyLight 488 conjugated Streptavidin (S000-41 lot 21097) 1:10000 for 30 min. Well was washed 3X in PBS.

Anti-Lactate Dehydrogenase Antibody - Western Blot. Western blot analysis shows Anti-Lactate Dehydrogenase antibody detects LDH in HeLa cell extracts. Reactivity with LDH is observed in the cytoplasmic fraction (CF) and little to no reactivity in the nuclear fraction (NF). ~30 ug was loaded per lane onto a 10% gel for SDS-PAGE. Comparison to a molecular weight marker (not shown) indicates a single band of ~36 kD corresponding to the expected molecular weight for the protein. The blot was incubated with a 1:1000 dilution of the antibody in 5% milk in TBST at room temperature followed by detection using standard procedures. Personal communication Kuldeep Patel.
