Specificity
SLP-76 (SH2 domain-containing leukocyte protein) is an adaptor molecule that is involved in T cell development and activation. Thymocytes from transgenic mice expressing a SLP-76 mutant with deletion of the amino terminus are unable to develop to CD4-CD8- stage. Mature T cells do develop in mice expressing SLP-76 with mutations in proline rich region or SH2 domain, but they fail to upregulate CD69 or proliferate appropriately in response to anti-CD3 stimulation. SLP-76 is cytosolic protein identified as phosphoprotein migrating at 76 kD in SDS-PAGE. Amino-terminal domain of SLP-76 containing several tyrosines is followed by PR (proline rich) region and an SH2 domain. SLP-76 associates with LAT via Gads or Grb 2. The PR region of SLP-76 constitutively associates with carboxy-terminal SH3 domain of Gads and P-1 segment of the PR region was also identified that associates with the SH3 domain of PLC-gamma1. The PR region therefore provides another pathway for PLC-gamma1 recruitment to the LAT signalling complex in T cells. The SH2 domain of SLP-76 has been identified as the region involved in binding the serine/threonine kinase HPK1. HPK1 may act as both a positive and a negative regulator by promoting the Jnk mitogen activated protein kinase (MAPK) pathway and inhibiting the pathway leading to AP-1 activation.
