Specificity and Use

KMT2A / MLL antibody was raised against a synthetic peptide mapping to a region between residues 720 and 780 of human myeloid/lymphoid or mixed-lineage leukemia 1 using the number given in Swiss-Prot entry Q03164 (GeneID 4297). The epitope is found in the N-terminal 300 kD fragment generated by proteolytic cleavage. The epitope is found in isoform 14P-18B of MLL1.
Specific for human MLL1 protein. Species cross-reactivity: mouse. Other species have not been tested.
Suitable for use in Western Blot, and Immunohistochemistry (paraffin-embedded sections).

Histone methyltransferase that plays an essential role in early development and hematopoiesis. Catalytic subunit of the MLL1/MLL complex, a multiprotein complex that mediates both methylation of 'Lys-4' of histone H3 (H3K4me) complex and acetylation of 'Lys-16' of histone H4 (H4K16ac). In the MLL1/MLL complex, it specifically mediates H3K4me, a specific tag for epigenetic transcriptional activation.

WB Analysis: Samples: Nuclear extract (50 ug for WT and KO; 10 ug for EF, EN and EC) from 293T cells that were transfected with MLL1 C-terminal 180 kD fragment (EC), 293T cells transfected with N-terminally truncated MLL1 fragment (EN; N300-tr), 293T cells transfected with MLL1 N-terminal 300 kD fragment (EF), MLL1 +/+ mouse embryonic stem cells (WT), and MLL1 -/- embryonic stem cells (KO). Antibody used at the indicated concentrations. Detection: One second exposure with chemiluminescence.
