WB Analysis: Samples: Nuclear extract (50 ug for WT and KO; 10 ug for EF, EN and EC) from 293T cells that were transfected with MLL1 C-terminal 180 kD fragment (EC), 293T cells transfected with N-terminally truncated MLL1 fragment (EN; N300-tr), 293T cells transfected with MLL1 N-terminal 300 kD fragment (EF), MLL1 +/+ mouse embryonic stem cells (WT), and MLL1 -/- embryonic stem cells (KO). Antibody used at the indicated concentrations. Detection: One second exposure with chemiluminescence.

Detection of Human C-terminal 180 kDa Fragment of MLL1 by Western Blot and Immunoprecipitation. Samples: Whole cell lysate from K562 (15 and 50 ug for WB; 1 mg for IP, 20% of IP loaded), 293T (T; 50 ug) and HeLa (H; 50 ug) cells. Antibodies: Affinity purified rabbit anti-MLL1 antibody used for WB at 0.4 ug/ml (A) and 1 ug/ml (B) and used for IP at 6 ug/mg lysate. The c-terminal 180 kDa Fragment of MLL1 was also immunoprecipitated by a prior lot of this antibody. Detection: Chemiluminescence with exposure times of 3 minutes (A) and 10 seconds (B).

KMT2A / MLL antibody was raised against mBP fusion protein corresponding to residues 3084-3959 of human mixed lineage leukemia (MLL).
Recognizes the C-terminal proteolytic fragment of human MLL (MLLC), Mr ~180kD. Species cross-reactivity: Human and mouse.
Suitable for use in Western Blot and Immunoprecipitation. Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and a chemiluminescence detection system. Immunoprecipitation: Reported by an independent laboratory to immunoprecipitate MLLC from nuclear extracts. The N-terminal fragment, MLLN (300kD), is also co-immunoprecipitated by this antibody.

About KMT2A / MLL
Histone methyltransferase that plays an essential role in early development and hematopoiesis. Catalytic subunit of the MLL1/MLL complex, a multiprotein complex that mediates both methylation of 'Lys-4' of histone H3 (H3K4me) complex and acetylation of 'Lys-16' of histone H4 (H4K16ac). In the MLL1/MLL complex, it specifically mediates H3K4me, a specific tag for epigenetic transcriptional activation. Q03164 NM_005933 NP_005924.2
