Specificity and Use

ITGB6 / Integrin Beta 6 antibody was raised against nS0-derived rhIntegrin beta 6.
Human Integrin beta 6 Immunogen: NS0-derived rhIntegrin beta 6 Ig Type: sheep IgG.
Western blot-This antibody can be used at 0.1-0.2 µg/ml with the appropriate secondary reagents to detect human Integrin beta 6. The detection limit for rhIntegrin beta 6 is approximately 1 ng/lane and 10 ng/lane under non-reducing and reducing conditions, respectively. Immunocytochemistry-This antibody has been used at a concentration of 10 µg/ml to detect human Integrin beta 6 in HT-29 human adenocarcinoma cells. Cells were fixed with PBS containing 4% paraformaldehyde for 20 minutes at room temperature and blocked with PBS containing 10% normal donkey serum, 0.1% Triton X-100, and. 1 % BSA for 45 minutes at room temperature. After blocking, cells were incubated with diluted primary antibody for 3 hours at room temperature followed by donkey anti-sheep lgG at room temperature for 1 hour. Between each step, cells were washed with PBS containing 0.1 % BSA. Flow cytometry-This antibody has been tested on HT-29 human adenocarcinoma cells for use in flow cytometry. Dilute this antibody to 50 µg/ml and add 10 ul of the diluted solution to 1-2.5 x 105 cells in a total reaction volume not exceeding 200 ul. The binding of unlabeled antibodies may be visualized by adding 10 ul of a 25 µg/ml stock solution of a secondary developing reagent such as anti-sheep lgG conjugated to a fluorochrome. Direct ELISA-This antibody can be used at 0.5-1.0 µg/ml with the appropriate secondary reagents to detect human Integrin beta 6. The detection limit for rhIntegrin beta 6 is approximately 0.1 ng/well. In this format, this antibody shows approximately. 35% cross-reactivity with rmIntegrin beta 6 and less than 5% cross-reactivity with rhIntegrin beta 3 and rhIntegrin beta 5.

ITGB6 / Integrin Beta 6 is a protein that is a member of the integrin superfamily. Members of this family are adhesion receptors that function in signaling from the extracellular matrix to the cell. Integrins are heterodimeric integral membrane proteins composed of an alpha chain and a beta chain. The encoded protein forms a dimer with an alpha v chain and this heterodimer can bind to ligands like fibronectin and transforming growth factor beta 1.
