Specificity and Use

Insulin antibody was raised against recombinant C-peptide construction.
No cross-reactivity with human, bovine, porcine and mouse/rat insulin. Affinity (Kd) >1 x 108 M all MAbs. All MAbs (except 2B7, 7E10 and 2A11) bind with equal efficiency to the human free C-peptide and C-peptide region in proinsulin molecules. Antigenic determinants identified by MAbs are in the three-dimensional structure generated by N- and C- terminal regions of C-peptide separated by beta-turn at position 47-50 of proinsulin. MAbs 2B7, 7E10 and 2A11 react only with rat C-peptide.

After removal of the precursor signal peptide, proinsulin is post-translationally cleaved into three peptides: the B chain and A chain peptides, which are covalently linked via two disulfide bonds to form insulin, and C-peptide. Binding of insulin to the insulin receptor (INSR) stimulates glucose uptake. A multitude of mutant alleles with phenotypic effects have been identified.
