Specificity and Use

IL17F antibody was raised against this purified antibody was prepared from whole rabbit serum produced by repeated immunizations with full length recombinant mouse IL17-F protein.
IL-17F antibody has been heated to 56°C for 30 minutes. In ELISA and other immunoreactive assays, this antibody will recognize both native and recombinant mouse IL17-F in cell supernatants and certain body fluids. A control of similarly diluted normal rabbit IgG is recommended.
This purified antibody has been tested for use in ELISA and western blotting. Reactivity is also expected in neutralizations, radioimmunoassay and immunohistochemistry. The endotoxin content is estimated to be <10 pg/ul by the LAL method. By western blot a band approximately 17.9 kD in size corresponding to mouse IL17-F protein is expected in the appropriate cell lysate or extract. IL-17F forms a disulfide linked homodimer. Specific conditions for reactivity should be optimized by the end user.

Anti-Mouse IL-17F Antibody - Western Blot. Western blot of anti-Mouse IL-17F antibody shows detection of a band ~18 kD in size corresponding to recombinant mouse IL-17F. Recombinant mouse IL-17F was loaded on to an SDS-PAGE gel at 0.25 ug and after separation was transferred to nitrocellulose. The membrane was blocked with 1% BSA in TBST for 30 min at RT, followed by incubation with primary antibody diluted 1:1000 in 1% BSA in TBST overnight at 4C. After washes, the blot was reacted with secondary antibody HRP Goat anti-Rabbit IgG antibody diluted 1:40000 in blocking buffer (MB-070) for 30 min at RT followed by reaction with FemtoMax Chemiluminescent substrate (p/n FEMTOMAX-110). Data was collected using Bio-Rad VersaDoc 4000 MP imaging system.
