Specificity and Use

HTR1A / 5-HT1A Receptor antibody was raised against synthetic peptide C-QPKKSVNGESGSRN from an internal region of human 5HT1A Receptor (NP_000515.2). Percent identity by BLAST analysis: Human, Chimpanzee, Gorilla, Orangutan, Gibbon, Marmoset (100%); Monkey (93%); Panda (86%).

Peptide ELISA: antibody detection limit dilution 1:16000. Western blot: Preliminary experiments gave bands at approx 37kD and 35kD in Human Brain (Hippocampus, Substantia nigra) lysates after 0.1 ug/ml antibody staining. Please note that currently we cannot find an explanation in the literature for the bands we observe given the calculated size of 46.1kD according to NP_000515.2. Both detected bands were successfully blocked by incubation with the immunizing peptide (and BLAST results with the immunizing peptide sequence did not identify any other proteins to explain the additional bands).

G-protein coupled receptor for 5-hydroxytryptamine (serotonin). Also functions as a receptor for various drugs and psychoactive substances. Ligand binding causes a conformation change that triggers signaling via guanine nucleotide-binding proteins (G proteins) and modulates the activity of down-stream effectors, such as adenylate cyclase. Beta-arrestin family members inhibit signaling via G proteins and mediate activation of alternative signaling pathways.
