Specificity and Use

HSPB1 / HSP27 antibody was raised against recombinant human Hsp27 (1-205aa) purified from E. coli.
Immunohistochemistry: LS-B2932 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B2932 was determined to be 10 ug/ml.

Heat shock protein 27 (Hsp27) also known as heat shock protein beta-1 (HSPB1) is a protein that in humans is encoded by the HSPB1 gene. Hsp27 is a chaperone of the sHsp (small heat shock protein) group among ubiquitin, a-crystallin, Hsp20 and others. The common functions of sHsps are chaperone activity, thermotolerance, inhibition of apoptosis, regulation of cell development, and cell differentiation. They also take part in signal transduction.

Cell lysates of HeLa (each 30 ug) were resolved by SDS-PAGE, transferred to PVDF membrane and probed with anti-human Hsp27 (1:1000). Proteins were visualized using a goat anti-mouse secondary antibody conjugated to HRP and an ECL detection system.
