Specificity and Use

Immunohistochemistry: LS-B1659 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B1659 was determined to be 15 ug/ml.

HNRNPK / hnRNP K belongs to the subfamily of ubiquitously expressed heterogeneous nuclear ribonucleoproteins (hnRNPs). The hnRNPs are RNA binding proteins and they complex with heterogeneous nuclear RNA (hnRNA). These proteins are associated with pre-mRNAs in the nucleus and appear to influence pre-mRNA processing and other aspects of mRNA metabolism and transport. While all of the hnRNPs are present in the nucleus, some seem to shuttle between the nucleus and the cytoplasm.

HeLa cell extracts were resolved by electrophoresis, transferred to nitrocellulose, and probed with anti-hnRNPK antibody (clone F45P9C7). Proteins were visualized using a goat anti-mouse-IgG secondary conjugated to HRP and chemiluminescence detection.
