Histone H4 antibody was raised against kLH-conjugated, synthetic peptide AGG[AcK]GG[AcK]GG[AcK]GG[AcK]GG[AcK]GGC related to amino acids 2 to 19 of Tetrahymena thermophila histone H2A.
Recognizes hyperacetylated histone H4, Mr 10kD (SDS-PAGE); tri-and tetra-acetylated histone H4 on acid-urea gels. May cross-react with other acetylated proteins. Species cross-reactivity: Human.
Suitable for use in Western Blot, Immunoprecipitation, Immunohistochemistry. Western Blot: 1:4000 (will detect hyperacetylated histone H4 in sodium butyrate treated, acid extracted HeLa nuclei resolved by SDS-PAGE) 1:3000 (will detect tri-and tetra-acetylated histone H4 from sodium butyrate treated, acid extracted HeLa nuclei resolved by acid-urea gel electrophoresis). Histones extracted from 0.5x1E6 cells were treated with 5 mM sodium butyrate for 6 hours, resolved by acid-urea gel electrophoresis and transferred to nitrocellulose. Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system. Immunohistochemistry: Reported to exclusively stain Tetrahymena macronuclei. Immunoprecipitation: Reported to immunoprecipitate transcriptionally active chromatin containing acetylated histone H4.
