Histone H4 antibody was raised against ovalbumin-conjugated, synthetic peptide (RGKGGAcKGLGKG-C) corresponding to aa3-13 of yeast Histone H4 acetylated on lysine 8, with a C-terminal cysteine added for conjugation purposes.
Recognizes acetyl-Histone H4 (Lys8), Mr 10kD. Does not detect Histone H4 from yeast containing a Histone H4 K8R substitution. Species cross-reactivity: Yeast and human. Broad species cross-reactivity is expected.
Suitable for use in Western Blot, Immunoprecipitation and Chromatin IP. Western Blot Analysis: 1:500-1:2000 dilution of this lot detected acetyl-Histone H4 (Lys8) in acid extracts from sodium butyrate treated HeLa cells. Immunoprecipitation: 5 ul of this lot immunoprecipitated acetyl Histone H4 from 1 mg of wild type whole cell yeast lysates but not from a yeast strain containing a Histone H4 K8R substitution. Chromatin IP: Has been reported by an independent laboratory to immunoprecipitate chromatin from wild type yeast strain but not from a yeast strain containing a Histone H4 K8R substitution.
