Histone H4 antibody was raised against ovalbumin-conjugated, synthetic peptide containing the sequence (. LG[acK]GG.) in which acK corresponds to acetyl-lysine 12 of yeast Histone H4.
Recognizes histone H4, Mr 10kD. Species cross-reactivity: Yeast and human. Broad species cross-reactivity expected.
Suitable for use in Peptide Inhibition, Peptide Dot Blot, Western Blot and Immunoprecipitation. Peptide Inhibition: 10 uM of a histone H4 peptide containing acetyl-lysine 12 abolished detection of histone H4 by anti-acetyl-Histone H4 (Lys12) in immunoblot analysis of HeLa acid extracts. Little to no signal reduction was observed with 10 uM of histone H4 peptides containing acetyl-lysine 5, acetyl-lysine 8 or acetyl-lysine 16. Peptide Dot Blot: A 1:1000 detects serial dilutions (25, 5, 1 and 0.2uM) of a histone H4 peptide containing acetyl-lysine 12 spotted onto PVDF, and not histone H4 peptides containing acetyl-lysine 5, acetyl-lysine 8 or acetyl-lysine 16. Western Blot: 1:3000 to 1:10000 detects acetyl-Histone H4 (Lys12) in acid extracted proteins from HeLa cells. The antibody did not detect unacetylated recombinant Histone H4. Immunoprecipitation: 2-6 ul immunoprecipitated the Ime2 promoter from wild type yeast chromatin and not from mutant yeast chromatin containing a histone H4 K12R mutation.
