H2AFX / H2AX antibody was raised against synthetic peptide from human H2AFX / H2AX.
Ser139
Amino acids surrounding Ser 139 of human H2AFX
Immunohistochemistry: LS-B1474 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B1474 was determined to be 10 ug/ml.

PBS (without Mg2+, Ca2+), pH 7.4, 150 mM sodium chloride, 0.02% sodium azide, 50% glycerol.

Variant histone H2A which replaces conventional H2A in a subset of nucleosomes. Nucleosomes wrap and compact DNA into chromatin, limiting DNA accessibility to the cellular machineries which require DNA as a template. Histones thereby play a central role in transcription regulation, DNA repair, DNA replication and chromosomal stability. DNA accessibility is regulated via a complex set of post-translational modifications of histones, also called histone code, and nucleosome remodeling.
