Specificity and Use

GSK3A / GSK3 Alpha antibody was raised against kLH conjugated, synthetic peptide CRARTS[pS]FAEPG, corresponding to amino acids 16-26 of the human GSK-3a.
pSer21
Recognizes phosphorylated human GSK-3a, Mr ~51kD, from cell lysates and phosphorylated recombinant GSK-3b. Does not recognize active dephosphorylated GSK-3a or GSK-3b. Predicted to cross-react with rat based on sequence homology (the immunizing sequence is identical in rat).
Suitable for use in Western Blot. Immunoprecipitation (not tested). Western Blot: 1:500-1:1000. RIPA lysates from HEK293 cells stimulated with 200 ng/ml of IGF1 for 5 minutes. Non-stimulated and IGF1-stimulated HEK293 cell lysates were resolved by electrophoresis, transferred to nitrocellulose and probed with anti-phospho-GSK3a (1:1000). Proteins, 51kD, were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system.

Constitutively active protein kinase that acts as a negative regulator in the hormonal control of glucose homeostasis, Wnt signaling and regulation of transcription factors and microtubules, by phosphorylating and inactivating glycogen synthase (GYS1 or GYS2), CTNNB1/beta-catenin, APC and AXIN1. Requires primed phosphorylation of the majority of its substrates. Contributes to insulin regulation of glycogen synthesis by phosphorylating and inhibiting GYS1 activity and hence glycogen synthesis.
