Immunohistochemical analysis of GLUR2 (pS880) staining in human brain formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP conjugated compact polymer system. DAB was used as the chromogen. The section was then counterstained with hematoxylin and mounted with DPX.

Specificity and Use

GRIA2 / GLUR2 antibody was raised against keyhole limpet hemocyanin conjugated synthetic peptide corresponding to an amino acid sequence within rat GluR2.
Recognizes glutamate receptor subunit 2 (GluR2), a component of the AMPA (alpha-amino-3-hydroxy-5-methyl-4-isoxalone propionic acid) group of ionotropic glutamate receptors, which play a key role at excitatory synapses, including synaptic transmission, stabilization and plasticity. GluR2 is possibly the most important of the AMPA receptor subunits, responsible for AMPA receptor rectifying properties, control of ion flow and in particular the influx of calcium. The majority of GluR2 in the CNS is expressed in the GluR2(R) form, containing a critical arginine residue (as opposed to a glutamine residue) in the Transmembrane region 2 (TM2) domain, thereby rendering native AMPA receptors impermeable to calcium.
Western Blot: LS-C43151 detects a band of approximately 100 kD in rat brain hippocampus cell lysates.

Glutamate receptors are the predominant excitatory neurotransmitter receptors in the mammalian brain and are activated in a variety of normal neurophysiologic processes. This gene product belongs to a family of glutamate receptors that are sensitive to alpha-amino-3-hydroxy-5-methyl-4-isoxazole propionate (AMPA), and function as ligand-activated cation channels. These channels are assembled from 4 related subunits, GRIA1-4.
