Immunohistochemical analysis of GRB10 staining in human breast cancer formalin fixed paraffin embedded tissue section. The section was pre-treated using heat mediated antigen retrieval with sodium citrate buffer (pH 6.0). The section was then incubated with the antibody at room temperature and detected using an HRP polymer system. DAB was used as the chromogen. The section was then counterstained with hematoxylin and mounted with DPX.

Specificity and Use

GRB10 antibody was raised against synthetic peptide corresponding to N-terminal residues of human GRB10 (KLH).
N-Terminus
Recognizes endogenous levels of human total GRB10 protein independent of phosphorylation at 76kD. Does not cross-react with GRB7 or GRB14.
Suitable for use in ELISA, Western Blot and Immunoprecipitation. Western Blot: 1:1000. Immunoprecipitation: 1:50.

Adapter protein which modulates coupling of a number of cell surface receptor kinases with specific signaling pathways. Binds to, and suppress signals from, activated receptors tyrosine kinases, including the insulin (INSR) and insulin-like growth factor (IGF1R) receptors. The inhibitory effect can be achieved by 2 mechanisms: interference with the signaling pathway and increased receptor degradation. Delays and reduces AKT1 phosphorylation in response to insulin stimulation.
