Specificity and Use

GPX4 / MCSP antibody was raised against synthetic peptide C-EEPLVIEKDLPHY from the C-terminus of human GPX4 (NP_002076.2; NP_001034937.1). Percent identity by BLAST analysis: Human, Gorilla, Orangutan, Gibbon, Monkey (100%); Goat, Hamster, Bovine (92%); Marmoset, Mouse, Rat, Pig, Opossum, Guinea pig (85%).

Human GPX4. This antibody is expected to recognise isoform A (NP_002076.2) and isoform C (NP_001034937.1).
Immunohistochemistry: LS-B3246 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B3246 was determined to be 2.5 ug/ml.

GPX4 / MCSP is a member of the glutathione peroxidase protein family. Glutathione peroxidase catalyzes the reduction of hydrogen peroxide, organic hydroperoxide, and lipid peroxides by reduced glutathione and functions in the protection of cells against oxidative damage. Human plasma glutathione peroxidase has been shown to be a selenium-containing enzyme and the UGA codon is translated into a selenocysteine.
