Specificity and Use

GPV / CD42d antibody was raised against human platelets.
The monoclonal antibody is directed against the 82 kD GP V antigen, which is expressed on human platelets. The monoclonal antibody reacts with human platelet. It has been shown that the glycoproteins GP V and GPIb-GP IX form a noncovalent complex in the platelet membrane. The monoclonal antibody does not react with lymphocytes, granulocytes, monocytes and erythrocytes. Antigen is expressed on human platelets and megakaryocytes. The antigen is absent or present in very low levels on platelets of patients with the Bernard-Soulier syndrome. Immunogen: human platelets.
Functional studies on human platelets. The monoclonal antibody does not inhibit the von Willebrand factor-mediated agglutination of fixed platelets induced by 1.25 mg/ml of ristocetin in the presence of human platelets. The antibody is useful for indirect immunofluorescence staining with analysis by FACS or fluorescence microscopy.

Human platelet glycoprotein V (GP5) is a part of the Ib-V-IX system of surface glycoproteins that constitute the receptor for von Willebrand factor (VWF; MIM 613160) and mediate the adhesion of platelets to injured vascular surfaces in the arterial circulation, a critical initiating event in hemostasis. The main portion of the receptor is a heterodimer composed of 2 polypeptide chains, an alpha chain (GP1BA; MIM 606672) and a beta chain (GP1BB; MIM 138720), that are linked by disulfide bonds.
