Specificity and Use

GPR75 antibody was raised against synthetic 19 amino acid peptide from 3rd cytoplasmic domain of human GPR75. Percent identity with other species by BLAST analysis: Human, Gorilla, Monkey (100%); Bovine, Pig (95%); Bat, Elephant, Panda, Rabbit, Horse (89%); Mouse, Rat, Xenopus (84%).

Human GPR75. BLAST analysis of the peptide immunogen showed no homology with other human proteins.
Immunohistochemistry: LS-A1591 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-A1591 was determined to be 20 ug/ml.

G protein-coupled receptor that is activated by the chemokine CCL5/RANTES. Probably coupled to heterotrimeric Gq proteins, it stimulates inositol trisphosphate production and calcium mobilization upon activation. Together with CCL5/RANTES, may play a role in neuron survival through activation of a downstream signaling pathway involving the PI3, Akt and MAP kinases. CCL5/RANTES may also regulate insulin secretion by pancreatic islet cells through activation of this receptor.
