Specificity and Use

GNAI1 / Gi antibody was raised against synthetic peptide (KNNLKDCGLF) corresponding to the C-terminal region shared by rat Gia-1 and Gia-2, coupled to KLH. Percent identity by BLAST analysis: Human, Gorilla, Orangutan, Gibbon, Monkey, Marmoset, Mouse, Rat, Hamster, Elephant, Panda, Bat, Bovine, Dog, Horse, Rabbit, Pig, Opossum, Guinea pig, Turkey, Chicken, Platypus, Lizard, Xenopus, Seabass, Catfish, Salmon, Stickleback, Medaka, Pufferfish, Zebrafish (100%).

Recognizes rat Gia-1 and Gia-2, Mr 40kD. Does not cross-react with Gia-3 or Goa. Species cross-reactivity: Human, bovine and mouse.
Suitable for use in Western Blot, Immunoprecipitation and Immunocytochemistry. Proteins were visualized using a goat anti-rabbit secondary antibody conjugated to HRP and a chemiluminescence detection system. Immunocytochemistry: 1:500 dilution gave positive immunostaining of mouse 3T3/A31 fibroblasts fixed for 3 minutes with ice-cold 100% acetone.

Guanine nucleotide-binding proteins (G proteins) are involved as modulators or transducers in various transmembrane signaling systems. The G(i) proteins are involved in hormonal regulation of adenylate cyclase: they inhibit the cyclase in response to beta-adrenergic stimuli. The inactive GDP-bound form prevents the association of RGS14 with centrosomes and is required for the translocation of RGS14 from the cytoplasm to the plasma membrane. May play a role in cell division.
