Specificity and Use

GLUD1/Glutamate Dehydrogenase antibody was raised against bovine liver glutamate dehydrogenase.
Recognizes Glutamate Dehydrogenase (GDH). Cross-reactivity is expected for mitochondrial and brain isoforms of GDH (GDH1 and GDH2). Assay by IEP resulted in a single precipitin arc against purified and partially purified bovine liver glutamate dehydrogenase. Species cross-reactivity: Most primate species (including green monkey, white gibbon, chimpanzee orangutan and gorilla), chicken, mouse, rat, canine and other mammals as well as Xenopus tropicalis, zebrafish, rainbow trout and Atlantic salmon is expected. Cross-reactivity with GDH from other tissues and species may occur.
Tested in IHC-P using formalin-fixed paraffin-embedded human tisseu samples.Suitable for Western Blot (Western and Dot Blot) and ELISA. ELISA: 1:4000-1:16000. Sandwich ELISA coat plates with 1 ug glutamate dehydrogenase and use peroxidase-conjugated goat anti-rabbit IgG. Western Blot: 1:1000-1:3000. Expect a band ~56kD corresponding to glutamate dehydrogenase monomer subunit in the appropriate cell or tissue extract.

GLUD1/Glutamate Dehydrogenase is glutamate dehydrogenase protein; a mitochondrial matrix enzyme that catalyzes the oxidative deamination of glutamate to alpha-ketoglutarate and ammonia. This enzyme has an important role in regulating amino acid induced insulin secretion and activating mutations in this gene are a common cause of congenital hyperinsulinism. This enzyme is allosterically activated by ADP and inhibited by GTP and ATP.
