Specificity and Use

GAD1 / GAD67 antibody was raised against synthetic peptide C-PDSPQRREKLHK from an internal region of human GAD1 / GAD67 (NP_000808.2). Percent identity by BLAST analysis: Human, Chimpanzee, Gorilla, Orangutan, Gibbon, Monkey (100%); Marmoset (92%); Mouse, Rat, Elephant, Panda, Bat, Bovine, Dog, Cat, Horse, Rabbit, Pig, Opossum (83%).

Human GAD1 / GAD67. This antibody is expected to recognize isoform GAD67. There is no cross-reactivity expected with GAD2.
Immunohistochemistry: LS-B3027 was validated for use in immunohistochemistry on a panel of 21 formalin-fixed, paraffin-embedded (FFPE) human tissues after heat induced antigen retrieval in pH 6.0 citrate buffer. After incubation with the primary antibody, slides were incubated with biotinylated secondary antibody, followed by alkaline phosphatase-streptavidin and chromogen. The stained slides were evaluated by a pathologist to confirm staining specificity. The optimal working concentration for LS-B3027 was determined to be 4 ug/ml. Peptide ELISA: antibody detection limit dilution 1:8000. Western Blot: Approx. 70kD band observed in Human Brain (Cerebellum) and in Mouse and Rat Brain lysates (calculated MW of 66.9kD according to Human NP_000808.2 and 66.6kD according to Mouse NP_032103.2). Recommended concentration: 0.3-1 ug/ml.

GAD1 / GAD67 is one of several forms of glutamic acid decarboxylase, identified as a major autoantigen in insulin-dependent diabetes. The enzyme encoded is responsible for catalyzing the production of gamma-aminobutyric acid from L-glutamic acid. A pathogenic role for this enzyme has been identified in the human pancreas since it has been identified as an autoantigen and an autoreactive T cell target in insulin-dependent diabetes. This gene may also play a role in the stiff man syndrome.
